Delayed Release of Ibuprofen By A Novel Solvent-less Dry Powder Coating

Abstract:#

A continuous solvent-less coating method is developed via twin screw hot melt extrusion to delay the release of fine ibuprofen (IBU) (50 μm) without using any liquid plasticizers, solvents, binders. The processing temperature is controlled at lower than that for the degradation of APIs/polymers. After coating process, particle size of wax coated samples is less than 200 μm, which is comparable to the other dry film coating method. The results show that 20 wt% of wax loading could fully covered the surface of IBU. The dissolution testing shows that t60% dissolution time of IBU increased from less than 1 minutes to more than 10 minutes. Korsmeyer-Peppas equation is used to explain and understand the release mechanism before and after coating. It is observed that Ksp is inversely proportional to the square of particle size and wax loading, which means wax coating reduce the effective surface during dissolution. This novel continuous solvent-less coating technique proved to be a very promising method in the control release of pharmaceuticals.
